Using Biotinylated myo-Inositol Hexakisphosphate to Investigate Inositol Pyrophosphate–Protein Interactions with Surface-Based Biosensors

چکیده

Inositol pyrophosphates (PP-InsPs) are highly phosphorylated molecules that have emerged as central nutrient messengers in eukaryotic organisms. They can bind to structurally diverse target proteins regulate biological functions, such protein–protein interactions. PP-InsPs strongly negatively charged and interact with basic surface patches proteins, making their quantitative biochemical analysis challenging. Here, we present the synthesis of biotinylated myo-inositol hexakisphosphates application plasmon resonance grating-coupled interferometry assays, enable rapid identification, validation, kinetic characterization InsP– PP-InsP–protein

Broad Spectrum Anticancer Activity of Myo-Inositol and Inositol Hexakisphosphate

Inositols (myo-inositol and inositol hexakisphosphate) exert a wide range of critical activities in both physiological and pathological settings. Deregulated inositol metabolism has been recorded in a number of diseases, including cancer, where inositol modulates different critical pathways. Inositols inhibit pRB phosphorylation, fostering the pRB/E2F complexes formation and blocking progressio...

Metabolism of inositol pentakisphosphate to inositol hexakisphosphate in Xenopus laevis oocytes.

The formation and metabolism of inositol pentakis-and hexakisphosphates (InsP5 and InsP6) were investigated in Xenopus laevis oocytes. After [3H]inositol injection, [3H]InsP5 and subsequently [3H]Insp6 increased progressively over 72 h. In intact oocytes, [3H]InsP5 was progressively converted to [3H]InsP6 from 6 to 72 h of incubation and was not metabolized to lower inositol phosphates. In cont...

Assessing the omnipotence of inositol hexakisphosphate.

This review assesses the authenticity of inositol hexakisphosphate (InsP(6)) being a wide-ranging regulator of many important cellular functions. Against a background in which the possible importance of localized InsP(6) metabolism is discussed, there is the facile explanation that InsP(6) is merely an "inactive" precursor for the diphosphorylated inositol phosphates. Indeed, many of the propos...

Interaction of myo-inositol hexakisphosphate with biogenic and synthetic polyamines.

Within all the eukaryotic cells myo-inositol phosphates (InsPs) are an important group of biomolecules that are potentially related to signaling functions. The most abundant member of this family in nature is InsP6 (phytate, L(12-) in its fully deprotonated form). The complicated chemical behavior of this molecule demands a great effort to understand its function in the cell medium. In this wor...

Mass changes of inositol 1,3,4,5,6-pentakisphosphate and inositol hexakisphosphate during cell cycle progression in rat thymocytes.

Changes in the cellular mass and the cellular concentration of the highly phosphorylated inositol derivatives inositol 1,3,4,5,6-pentakisphosphate (Ins(1,3,4,5,6)P5) and inositol hexakisphosphate (InsP6) were observed during a complete cell cycle of proliferating rat thymocytes. Inositol polyphosphates were determined by a recently developed anion-exchange HPLC/post-column complexometric dye sy...
